Biosynthesis of plasma lipoproteins. Incorporation of 14C-glucosamine by cells and subcellular fractions of rat liver.

چکیده
The incorporation of glucosamlneJ4C into plasma lipoproteins has been measured in the intact rat, in liver slices, and in the liver microsome fraction. The labeling of lipoprotein was studied in microsome subfractions, both in viva with glucosammeJ4C and in vitro with UDP-N-acetylglucosamineW. After isolation of lipoproteins with the aid of carrier plasma, labeled lipoproteins were found in smooth endoplasmic reticulum and Golgi membranes, but not in rough endoplasmic reticulum or ribosomes. Approximately 100 times as much smooth membrane protein as Golgi membrane protein was required to isolate the same total amount of labeled lipoprotein, in uivo as well as in vitro. High density lipoproteins had 3 times as much total label as low density lipoproteins, which may be related to the larger amount of the former molecules found in plasma. It is concluded that carbohydrate is attached to the lipoprotein after it has left the ribosome and that the Golgi apparatus plays a major role in this process.

The Biosynthesis of Glycoproteins III. GLUCOSAMINE INTERMEDIATES IN PLASMA GLYCOPROTEIN SYNTHESIS IN LIVERS
It was previously shown that the particulate fraction of liver was the site of incorporation of glucosamine-l-14C into rat plasma glycoproteins (1). In continuing these studies, we wished to determine whether this incorporation was dependent on the synthesis of protein de nova. Since puromycin inhibits protein biosynthesis (2-5), we have studied the effect of this drug on the incorporation of g...

متن کاملBiosynthesis of Glycoproteins
The presence of carbohydrate in many plasma and tissue proteins is well established (1, 2). The glycoproteins thus far studied contain one or several oligosaccharide units linked to a peptide chain by uncharacterized linkages. Little is yet known about the biosynthetic processes by which the oligosaccharide units are assembled and attached to the polypeptide chain. In order to approach these pr...
متن کاملThe Initial Subcellular Site of Incorporation of Hexoses into Liver Protein.
Since glycoproteins contain both peptide chains and covalently linked carbohydrates, challenging questions regarding their mode of synthesis are where and how cells incorporate nonprotein molecules such as carbohydrates to form glycoproteins. The present study was undertaken to establish the subcellular site of incorporation of hexoses into liver protein. In achieving this end, information was ...
متن کاملDolichol metabolism in rat liver. Determination of the subcellular distribution of dolichyl phosphate and its site and rate of de novo biosynthesis.
Two independent experiments indicated that the major subcellular site of de novo dolichyl phosphate biosynthesis in rat liver is in the microsomes. First, when purified subcellular fractions were assayed for long chain prenyltransferase activity, the profile obtained indicated exclusively a microsomal localization. Second, when liver slices were incubated 60 min with [14C]acetate and subcellula...
